• Description:
  Rytm theta jest jednym z podstawowych, a zarazem najlepiej zsynchronizowanych wzorców elektroencefalograficznych, który występuje w mózgach ssaków. U szczurów rytm ten rejestrowany jest w postaci prawie sinusoidalnych wysokoamplitudowych (1-2 mV) oscylacji o częstotliwościach od 3 do 12 Hz. Rytm theta występuje w licznych obszarach mózgu: korze zakrętu obręczy, korze śródwęchowej, korze nowej. Powszechnie uważa się jednak, że główną strukturą przejawiającą oscylacje theta jest formacja hipokampa (HPC). Obecność rytmu w HPC jest następstwem aktywacji tzw. wstępującego układu synchronizującego. System ten bierze swój początek w jądrach pnia mózgu: jądrze konarowo-mostowym nakrywki i przednim jądrze siatkowatym mostu, skąd toniczne pobudzenie wysyłane jest do obszaru tylnego podwzgórza. Na tym poziomie, główne jądra tylnego podwzgórza (PH) oraz jądro nadsuteczkowate (SuM) uczestniczą w przekazywaniu informacji pobudzającej do kolejnego obszaru, czyli kompleksu przegrody przyśrodkowej (MS/vDBB). Kompleks ten składa się z jąder przegrody przyśrodkowej oraz jądra pasma przekątnego Broca i uczestniczy nie tylko w przekazywaniu pobudzającej informacji ale również częściowo w kodowaniu właściwej częstotliwości rytmu theta. Pobudzenie o rytmicznym charakterze pochodzące z MS/vDBB dociera do HPC oraz przyległych struktur limbicznych, gdzie rytm theta jest generowany i skąd jest rejestrowany z najwyższą amplitudą. Dowiedziono, że rytm theta można również skutecznie rejestrować z odnerwionych preparatów mózgowych (skrawków) np. formacji hipokampa utrzymywanych w warunkach pozaustrojowych. W tych warunkach, rejestruje się podtyp II rytmu theta, w postaci oscylacji w paśmie częstotliwości od 4 do 9 Hz. Obszar tylnego podwzgórza nie był nigdy dokładnie badany pod względem jego zdolności do genezy rytmu, choć wiadomo, że w niektórych stanach behawioralnych uczestniczy w kontroli częstotliwości hipokampalnego rytmu theta. W niniejszej rozprawie doktorskiej po raz pierwszy zaprezentowane zostały wyniki rejestracji rytmu theta z preparatów tylnego podwzgórza utrzymywanych w warunkach in vitro. Dowiedziono, że w określonych warunkach, obszar ten jest zdolny do lokalnej rytmogenezy. W zaprezentowanym cyklu badawczym dokonano też mapowania lokalizacji rytmu theta w wybranych jądrach tylnego podwzgórza. W kolejnych doświadczeniach badano udział synaps elektrycznych (GJs) w genezie lokalnego, cholinergicznego rytmu theta w skrawkach tylnego podwzgórza. Na podstawie uzyskanych wyników należy przypuszczać, że GJs nie są zaangażowane w proces genezy rytmu theta w jadrach SuM i PH. W toku przeprowadzonych eksperymentów zauważono jednak poprawę średnich parametrów rytmu po podaniu blokera GJs – karbenoksolonu. Przypuszczalnie, efekt ten jest następstwem działania niezwiązanego z elektrycznym przekaźnictwem synaptycznym. W ramach badań do rozprawy doktorskiej dokonano również zewnątrzkomórkowych rejestracji aktywności neuronów w preparatach tylnego podwzgórza. Neurony klasyfikowane były na podstawie związku ich wzorca wyładowań z jednocześnie rejestrowanym lokalnym rytmem theta, oraz na podstawie istniejącej klasyfikacji komórek związanych z rytmem theta. Wyniki uzyskane w opisywanym cyklu badawczym potwierdzają istnienie neuronów theta-zależnych również w odnerwionych preparatach tylnego podwzgórza, tym samym dowodząc uniwersalności wcześniej opisanej klasyfikacji tych komórek. Ostatnim realizowanym cyklem badawczym była weryfikacja udziału układu oreksynergicznego w genezie lokalnego rytmu theta w jądrach tylnego podwzgórza. Oreksyny podawane samodzielnie nie wywoływały oscylacji w paśmie theta. Zaobserwowano jednak pozytywny efekt modulacyjny cholinergicznie indukowanego rytmu theta w wyniku zastosowania oreksyny B. Wyniki opisane w niniejszej rozprawie doktorskiej dowodzą, iż skrawki obszaru tylnego podwzgórza, w określonych warunkach, są zdolne do lokalnych procesów neuronalnej synchronizacji oraz genezy rytmu theta. Model wykorzystujący te skrawki jest użyteczny do badań neurofarmakologicznych oraz rejestracji aktywności komórkowej. Ponadto, uzyskane wyniki zdają się potwierdzać sugerowaną rolę jąder tylnego podwzgórza jako jednego z modulatorów częstotliwości hipokampalnego rytmu theta na drodze wstępującego systemu synchronizującego.;
  Theta rhythm is one of the best synchronized electroencephalographic (EEG) patterns that is present in mammalian brains. In rats, theta rhythm field potentials consist of high amplitude (1-2 mV) and almost sinusoidal oscillations in a narrow frequency band ranging from 3 to 12 Hz. Multiple brain areas were found capable of producing theta rhythm: the cingulate cortex, the entorhinal cortex, the neocortex. However, it is widely accepted that the most prominent brain structure in theta generation is the hippocampal formation (HPC). It is known that the appearance of HPC theta is a result of the ascending synchronizing pathway activation. This system begins in the pons with the nucleus reticularis pontis oralis and the pedunculopontine tegmental nucleus where a tonic drive is produced. These nuclei project to the posterior hypothalamic area – specifically, the supramammillary nucleus (SuM) and the primary posterior hypothalamic nuclei (PH). They in turn project to the medial septal complex (MS/vDBB) made up of medial septal nuclei and the diagonal band of Broca nucleus. Not only does the MS/vDBB provide an excitatory drive to the HPC but in some circumstances it also programs the frequency of theta rhythm. Finally, the excitatory rhythmic input reaches the HPC and other limbic structures where theta rhythm is generated with great amplitude. It has been previously established that theta rhythm can also be studied in in vitro conditions with the use of deafferented brain slices of a number of brain areas including the HPC. In these conditions, type II theta rhythm subtype can be recorded and it consists of a narrower frequency range oscillations (4-9 Hz). The posterior hypothalamic nuclei had never been studied extensively regarding their ability to produce local theta rhythm. However, it is known that they participate in the process of HPC theta frequency programing at least in some behavioral states. The current study provides evidence that the SuM and PH nuclei are in fact capable of generating local theta filed potentials in in vitro conditions. Additionally, mapping studies have been carried out in order to examine the probability of recording theta in different locations among mentioned nuclei. Examining the role of electrical synapses (GJs) was the next step in understanding the mechanism of rhythmogenesis in the posterior hypothalamic 15 slices. Obtained data suggests that GJs are not involved in the generation of local theta rhythm in the SuM and PH nuclei. On the other hand, a significant improvement in mean parameters of theta oscillations was observed after the administration of carbenoxolone, a GJs blocker. Experiments have been performed in order to examine the mechanism of this alteration and the results seem to confirm that this effect is not mediated by electrical synapses. During the next experiments, neuronal activity has been examined in regard to theta field activity in posterior hypothalamic slices. Recorded single neurons have been classified based on their relation to theta rhythm with the use of an existing classification of theta-related cells. For the first time, theta-related cells activity recorded from posterior hypothalamic slice preparations was observed and the cells were classified based on their relation to local theta oscillations. On the basis of the obtained results it is suggested that the posterior hypothalamic nuclei share a similar theta generation mechanism at a cellular level with the HPC. The final aim of this study was to investigate the involvement of orexinergic neurotransmitter system in the genesis of local theta field potentials in posterior hypothalamic preparations. It has been previously discovered that orexin administration in the HPC (both in vitro and in vivo) leads to well-synchronized theta rhythm generation. Analogous experiments have been carried out using in vitro posterior hypothalamic preparations. Orexin A or orexin B administration was not capable alone to induce theta oscillations in PH or SuM nuclei. However, a significant increase in mean amplitudes of cholinergically induced theta rhythm was observed after the administration of orexin B. This result suggests a modulatory role of orexin B in the process of cholinergic theta generation in the posterior hypothalamic preparations maintained in vitro. The presented results show for the first time that posterior hypothalamic slice preparations are capable of generation local cholinergic theta rhythm. It is proposed that posterior hypothalamic slices are a useful model for future neuropharmacological studies and single neuron activity recordings. Taken together, obtained results seem to underline the importance of posterior hypothalamic nuclei in the process of programming the frequency of hippocampal theta rhythm.
